Injection is required — thymulin is a 34-amino-acid peptide subject to complete proteolytic degradation in the gastrointestinal tract, with oral bioavailability effectively zero. Published research protocols use subcutaneous or intraperitoneal injection to achieve systemic circulation and thymic tissue distribution. Oral administration would not deliver intact peptide to target tissues and cannot replicate the thymic restoration effects documented in peer-reviewed studies using parenteral routes.
Essential controls include: (1) vehicle-only control group receiving bacteriostatic water or PBS at the same injection schedule to account for handling stress and injection trauma, (2) zinc-supplemented control group to isolate thymulin-specific effects from zinc availability, (3) positive control using established immunomodulatory compounds if comparing thymulin to alternative interventions, and (4) age-matched cohorts to control for baseline thymic function variance. Flow cytometry analysis of thymic populations should include isotype controls and fluorescence-minus-one (FMO) controls to ensure gate specificity for CD4, CD8, and activation markers.
